Harry A. Hoffner, Jr. wurde in Jacksonville (Florida) als jüngstes Kind von Harry A. Hoffner und Madaline Wolford Hoffner geboren. Seine schulische Laufbahn vollzog er in seiner Heimatstadt und in Pottstown (Pennsylvania), wo er von 1950 bis 1952 "The Hill School" (a university preparatory school) besuchte.

Im Jahre 1952 betritt Hoffner die "Princeton University" im Bundesstaat New Jersey mit dem Schwerpunkt Europäische Zivilisationen und Geschichte. Im Jahre 1956 schloss er mit dem Bachelor of Arts (Cum laude) in Princeton ab.

Im Sommer 1952 übersiedelte er von New Jersey nach Texas, um am "Dallas Theological Seminary" intensiver dort mit dem Hebräischen, der Bibel Exegese und Christlicher Theologie zu befassen, ein Gebiet, das ihn in Princeton zu interessieren begann und schloss mit dem Master of Theology (Highest Honors) ab.

Harry A. Hoffner, jr. vertiefte seine Studien ab 1960 an der "Brandeis University" in der Umgebung Bostons und ging zu Cyrus Herzl Gordon (1908 –2001), einer der führenden amerikanischen Experten für die Sprachen des Mittelmeeraumes. In Brandeis studierte er die altorientalische Sprachen und das klassische Arabisch. Seine altorientalistischen Grundlagen erhielt Hoffner hier, unter anderen das Akkadisch, Ugaritisch, Ägyptisch, und Arabisch. Während dieser Zeit erkannte Hoffner die kulturhistorische Bedeutung der Hethiter so dass er das Hethitische im Selbststudium anhand Friedrichs Hethitisches Elementarbuch erlernte.

1963 promovierte er bei Gordon mit der Arbeit The Laws of the Hittites  In diesem Jahr legte Harry A. Hoffner jr. auch den Grundstein zu seinem später am „Oriental Institute" in Chicago, gemeinsam Hans Gustav Güterbock, begonnenen Chicago HIttite Dictionnary, in dem er umfangreiche lexikalische Sammlungen anzufertigen begann. Ein hethitischer Thesaurus, der die Lemmata mit ihren Belegstellen und in ihren Kontexten aufführte war seit Beginn der Hethitologie ein Desideratum. Wie dem Vorwort der ersten Lieferung des CHD zu entnehmen ist, hatte Hoffner bis 1973 bereits 65% der publizierten Texte verzettelt und lexikalisch aufgenommen und somit die Voraussetzungen geschaffen, ein solches großes Projekt wie das CHD zu starten.

Von 1964 bis 1969 hatte Hoffner die Position eines Assistant Professor of Anatolian Studies an der Brandeis University inne, und dort waren ihm nach Ausweis seiner an Titeln so zahlreichen Publikationsliste, ausgesprochen produktive Jahre gewährt. Neben der schon erwähnten Erfassung des hethitischen Lexikons, publizierte Hoffner in dieser Periode zahlreiche Beiträge zu lexikalischen und syntaktischen aber auch religionsgeschichtlichen Problemen des Hethitischen. 1967 erschien als sichtbares Ergebnis seiner intensiven lexikalischen Studien An English-Hittite Glossary.

Im Jahre 1969 wurde Hoffner als Associate Professor of Hittitology an die "Yale University" (Connecticut) berufen. Während dieser Zeit entstand sein fundamentales Werk Alimenta Hethaeorum, das in methodisch beeindruckender Weise, die in den hethitischen Texten genannten Nahrungsmittel identifiziert. Hoffner geht zunächst vom landwirtschaftlichen Jahreskreislaufs Anatoliens aus und definiert die möglichen Anbaumöglichkeiten für die entsprechenden Jahreszeiten und die verschiedenen Belegmöglichkeiten. In das klare und überzeugende Geflecht der Argumentation werden die Ergebnisse der archäologischen Forschung eingeflochten. Die Kapitel über die verschiedenen Brote (NINDA) und deren Weiterverabeitungsprodukte, die vor allem in der Ritualliteratur und Festbeschreibungen belegt sind, sind eine eingehende Studie des schwierigen Genres der realkundlichen Bestimmung altorientalischer Fauna und Floranamen.

Im Herbst 1972 während eines Besuchs bei Heinrich Otten in Marburg und der Arbeit mit Hilfe des dortigen Archivs an der Fertigstellung seines Buch Alimenta Hethaeorum, erfuhr Hoffner, dass das Vorhaben Heinrich Ottens zur Herausgabe eines umfassenden Thesaurus Hethiticus sich noch für Jahre verzögern würde.

1974 wurde Hoffner als Professor of Hittite an die Universität Chicago und ihr berühmtes "Oriental Institute"  berufen. Hier traf er auf Hans Gustav Güterbock, der seit Jahrzehnten die Hethitologie an der Spitze der Forschung vorantrieb und über große Erfahrung in der lexikalischen Forschung verfügte. Die Sammlungen Hoffners und die Erfahrung Güterbocks ergänzten sich und mit einer Förderung des National Endowment for the Humanities begann das Chicago Hittite Dictionary Gestalt anzunehmen, ausgehend von dem Wunsch, dass Studenten und Forscher der englischsprachigen Welt ein Wörterbuch in ihrer Sprache haben sollten. Begonnen wurde mit dem Buchstaben "L", um das parallel in München von Annelies Kammenhuber in Arbeit befindliche Hethtitische Wörterbuch, welches mit dem Buchstaben "A" begonnen hatte, zu ergänzen. Der erste Band des CHD erschien 1979 und wird bis heute in Chicago fortgesetzt. Seit dem Jahre 2000 gibt es das CHD auch in einer elektronischen Form (Abk. eCHD), an der Harry A. Hoffner ebenfalls als Co-Direktor mitwirkt

1997 erschien aus der Feder Hoffners eine neue Ausgabe der hethitischen Gesetze, The Hittite Laws. A critical edition, in welcher er diese für die Hethitologie so wichtige Textgruppe einer umfassenden (362 Seiten) Neubearbeitung unterzog und damit nach fast vierzig Jahren die Arbeit Johannes Friedrichs von 1959 ersetzte.

Im Jahre 2008 wird das Werk Grammar of the Hittite Language, welches in Zusammenarbeit mit H.C. Melchert entstanden ist, erscheinen und auch auf diesem Gebiet der Hethitologie die gewünschte Aktualisierung des Wissens vornehmen. Angesichts dieses Oeuvres verbindet die wissenschaftliche Welt dankbar mit dem Namen Harry A. Hoffner jr. die Genese der für die Philologie des Hethitischen so grundlegenden Werkzeuge, allen voran das CHD, das über viele Jahrzehnte nun schon die Wissenschaft von den Hethitern begleitet und ihr Vorankommen fördert.

Seit dem Jahre 2000 ist Harry A. Hoffner emeritiert und lebt in Chicago. Er ist Vater von drei Kindern und noch heute Senior Editor des Chicago Hittite Dictionary. Anlässlich seines 65.
